Enhancement of immune cellular agglutination by use of an avidin-biotin system.

چکیده
Classically, one can increase the titer of an agglutinating or first antibody with an antiglobulin or second antibody. We have used an avidin-biotin system in place of antiglobulin to similarly extend the agglutination by an initial anticellular antibody. Erythrocytes were agglutinated by adding in succession, caproylamidobiotin-antibody, avidin, and extender (caproylamidobiotin-macromolecule). The macromolecules evaluated as extenders, in order of increasing potency, were fibrinogen, albumin, succinylated polylysine, and ribonuclease A. From comparative testing, we found that antiglobulin raised the titer of antibody from 2560 to 20,480, and the avidin-biotin tool raised the titer of caproylamidobiotin-antibody from 2560 to 10,240 without extender and to 81,820 with an extender of caproylamidobiotin-ribonuclease. Thus noncovalent extenders add to the capability of the avidin-biotin system to facilitate and substitute for an antibody.

Avidin-biotin latex agglutination assay for detection of antibodies to viral antigens.
A new method for attaching antigens to latex by an avidin-biotin technique is described. The procedure permits control of the amount of antigen attached to the latex and eliminates the need for highly purified antigens and destructive bridging chemicals. The avidin-biotin latex agglutination assay is a simple, rapid test well suited to detection of viral antibody. The sensitivity and specificit...
متن کاملLabel-free amplified bioaffinity detection using terahertz wave technology.
A new affinity biosensor based on pulsed terahertz (THz) wave technology has been used to monitor binding between biotin and avidin molecules. Amplified detection of avidin-biotin binding is obtained on supported membranes composed of biotin layers on quartz surface, which is modified with octadecanol. Agarose particles are conjugated with avidin and then applied to biotin, which is already bou...
متن کاملAvidin Attachment to Biotinylated Erythrocytes Induces Homologous Lysis Via the Alternative Pathway
Noncovalent attachment of avidin to the membrane of prebiotinylated red blood cells (RBCs) induces lysis via the alternative pathway of complement (APC). Lysis is not speciesdependent; RBCs from humans, rabbits, rats, and sheep were lysed with both autologous and all heterologous sera. Both biotinylated and native cells were not lysed. Lysis was observed at an avidin surface density of about 10...

Angiogenic, that is, newly formed, blood vessels play an important role in tumor growth and metastasis and are a potential target for tumor treatment. In previous studies, the alpha(v)beta(3) integrin, which is strongly expressed in angiogenic vessels, has been used as a target for Arg-Gly-Asp (RGD)-functionalized nanoparticulate contrast agents for magnetic resonance imaging-based visualizatio...
